Right after the events of Duke Nukem II, as Duke was returning to Earth in hopes of a vacation his ship was shot down by unknown hostiles. Upon landing, he finds out that Los Angeles has been overrun with aliens who mutated the LAPD. With his vacation plans ruined, Duke embarks on a mission to annihilate all aliens and free the city - again.
